在该研究中,根据电荷特性,酪蛋白水解产物分为4个部分。在模拟胃肠道和Caco-2细胞吸收之后,研究了肽被吸附物的细胞内抗氧化特性和LDL氧化抑制特性。与负电荷的肽片段相比,正电荷肽片段的肽被吸附物有着较高的细胞内抗氧化特性和LDL氧化抑制特性。通过超高效液相色谱电喷雾四级杆飞行时间串联质鉴定正电荷片段的消化物和被吸附物的肽序列。与氨基酸序列相比较,鉴定了35个完整的吸附肽。通过合成3个完整的吸附肽,被证实是完整的吸附肽,其能显著抑制LDL氧化。全部正电荷肽可能在抑制LDL氧化上发挥重要的作用。完整的吸附肽可能有体内真正的抗氧化效率。
完整的吸附肽的鉴定可能支持了正电荷肽的使用(作为天然抗氧化剂),以预防人动脉硬化。

Resistance to gastrointestinal (GI) digestion and absorption is necessary for bioactive peptides to exert their biological activities.
In this study, casein hydrolysate was separated into four fractions according to the charge property. After simulated GI digestion and Caco-2 cell absorption, the cellular antioxidant activity (CAA) and inhibition of LDL oxidation of peptide absorbates (PA) were investigated. The PA of positively charged fractions exhibited higher CAA and inhibition of LDL oxidation than negatively charged fractions. The peptide sequences from the digest and PA of the positively charged fractions were identified using UPLC-ESI-qTOF-MS. By comparing the amino acid sequences, 35 intact absorbed peptides (IAPs) were identified. Three IAPs were synthesized and all were verified to be IAPs, which significantly inhibited LDL oxidation. The overall positive charge property of peptides may play an important role in their ability to inhibit LDL oxidation, and IAPs may have real antioxidative efficacy in vivo.
Identification of IAPs should support the use of positively charged peptides as natural antioxidants for preventing human atherosclerosis.
